When Pratt was 19 years old, he waited tables at Bubba Gump Company restaurant in Maui, when actress and director Rae Dawn Chong discovered him. In her debut as a director, she portrayed him in the short horror film Cursed Part 3, which was shot in Los Angeles. On the television show Everwood, Pratt's first regular television appearance was as Harold Brighton "Bright" Abbott. Since Everwood's demise, he joined The O.C. cast. Winchester "Ché" Cook, a female activist, is back for his fourth season. He was also starring in the 2008 adventure film Wanted, in which James McAvoy attacks him with a keyboard.

Pratt debuted Andy Dwyer on the NBC comedy series Parks and Recreation in 2009. Producers loved Pratt so much that they asked him to become a series regular, even though he was supposed to be a temporary character. It was his breakthrough role in this regard.

In the 2011 film Moneyball, Pratt portrayed Oakland Athletics first baseman/catcher Scott Hatteberg. He was initially told that he was too fat to play Hatteberg because he had gained 40 pounds (18 kg), which Pratt attributed to his then-girlfriend, actress Anna Faris' cooking. He worked out every day to see if the role was cast, losing a total of 30 pounds (14 kg). He felt he had lost enough and sent a snapshot of himself to the casting director, who was able to capture the role. Pratt was portrayed as young and somewhat immature characters before the game of Moneyball was announced. He starred in Moneyball as a father and dejected baseball player who once feared his time was over and had to learn a new defensive position.

In September 2021, it was announced that Pratt would be voicing Nintendo mascot Mario in The Super Mario Bros. film based on the same name. It was revealed in November 2021 that he would be voicing the titular cat of the forthcoming animated film Garfield, directed by Mark Dindal from a script by David Reynolds. He was announced in June 2022 as a co-star in The Electric State alongside Millie Bobby Brown.
