Hillary Scott attended Middle Tennessee State University after high school. During her time at MTSU, Scott collaborated with singer-songwriter Victoria Shaw and gained some fame, but she was unable to secure a deal with a major label. She applied for American Idol twice before falling out of the judges' round in both cases, but she did not make it to the judge round. In May 2006, she met future bandmates Charles Kelley and later, Dave Haywood. She spotted Kelley in a Nashville bar and recognized him from his MySpace page, where she had been listening to his music. She introduced herself to Kelley and praised him on his music, which culminated in him deciding that they should get together with Haywood to write songs. "All We'll Ever Need" was the first song they wrote. The song was written for her to perform; she begged Kelley that it should be a duet, which resulted in Lady A's appearance, later known as Lady Antebellum. The group first appeared together in August 2006 and then signed a landmark deal with Capitol Nashville in 2007. Scott claims that being lead singer of Lady A rather than being a solo artist reduces the pressure on her on stage.

In 2006, Scott formed Lady Antebellum, co-lead singer Charles Kelley and backup singer Dave Haywood, who performed in the country music band. On October 2, 2007, they debuted their debut single, "Love Don't Live Here," on Billboard's Hot Country Songs chart for the first year, peaking at No. 62. In early 2008, there were three children in the country's northeast. Their debut album was released on April 15, 2008, and they debuted at No. 1 on April 15, 2008. 1 on the Top Country Albums chart. They launched "Just a Kiss" as the first single from their third studio album, Own the Night, on September 13, 2011, their fifth top-one hit, and their third consecutive number one hit in May 2011. The Royal Institute of British Architects had "Just a Kiss" rated as Platinum. It was on the Hot 100 for forty-two weeks. The second single, "We Owned the Night," became a number one hit and was subsequently named Gold. The third single "Dancin' Away with My Heart" was also awarded Gold and peaked at number two. As the fourth and final single from the album, "Wanted You More" was released. It was their first single since "Lookin' For a Good Time" fell out of the top ten. The band supported the album on their second headlining tour, the Own the Night Tour, which was also their first arena and world tour.

The band's first Christmas album was released on October 22, 2012, on This Winter's Night. Country radio and AC radio broadcast one of the album's "A Holly Jolly Christmas" track "A Holly Jolly Christmas." The single's music video was made.

The group began working on their fifth studio right after the publication of On This Winter's Night. Golden was first released on May 7, 2013. "Downtown" was the album's first single and ranked second on Country radio and first on the US Country Airplay chart. It was rated platinum by the Royal Institute of British Architects (RIAA). "Goodbye Town" was the second single on the album. The third single "Compass" was included in the deluxe version of Golden released in late 2013, which also included the third single "Compass." The band announced on June 11, 2020, during the George Floyd demonstrations in order to avoid racial connotations with Civil War history, as the original name was based on the Antebellum South. "We're deeply sorry for the pain" that has caused and for anyone who has been injured, unseen, or unvalued," the band posted on social media.

Scott was reportedly working on a gospel album in November 2015. She published a handwritten note about the campaign on April 22, 2016. "Hillary Scott & the Scott Family" revealed in a note in which she expressed her excitement about it, Love Remains and more specifically introduced "Hillary Scott & the Scott Family." "Well, I wanted to make this album to show more of who I am," she says. These songs, as well as others, are the cries of my heart. I've been singing songs like this for the rest of my life, whether it be in church, with family and friends assembled around with guitars on my screened porch, rocking my baby girl to sleep, or holding the hand of a dying parent." The album features original work penned by Scott as well as gospel guidelines, and was produced by Ricky Skaggs. Linda Davis and Lang Scott, Scott's parents, as well as younger sister Rylee appear on the album. On April 22, 2016, the lead single "Thy Will" was released.

Love Remains debuted on Billboard 200 at number 7 on the Billboard 200. Love Remains received Best Contemporary Christian Album and "Thy Will" received Best Contemporary Christian Music Performance/Song at the 59th Annual Grammy Awards.

Scott's other projects include singing background vocals on Luke Bryan's "Do I," which fellow Lady A bandmates, Charles Kelley and Dave Haywood wrote alongside Bryan. In April 2009, the song debuted on radio and peaked at No. 28. In December of this year, Billboard's Hot Country Songs chart placed 2 on the Billboard Hot Country Songs chart.

"Christmas Tonight" was Scott's duet with Dave Barnes, and she appeared on "Out of Goodbyes" with Kelley and Haywood, as Lady Antebellum. "Christmas Tonight" was a single off Barnes' 2010 holiday album "Mega Merry Christmas" and peaked at No. 1 on the charts. On New Year's Day, 2011, there are 9 on the Adult Contemporary chart. On Maroon 5's 2010 album, Hands All Over, "Out of Goodbyes," was included, but it was never released as a single.

In addition, Scott has written songs for other country artists, including Sara Evans' "A Little Bit More" and Blake Shelton's "Suffocating." Scott appeared on "A Little Bit Stronger" on harmony vocals and was announced in September 2010 as the lead off single on Evans' sixth studio album Stronger. The song was a hit in the 2010 film, Country Strong, and it topped the Hot Country Songs chart for two weeks in May 2011, becoming Scott's first-ever Number One hit by another artist. Scott with Shelton's ex-wife Miranda Lambert, and Shelton's All About Tonight recorded "suffocating" on his 2010 album, All About Tonight.

Scott served as a mentor for "Team Adam Levine" on season 4 of The Voice. In 2016, she hosted Gospel Hour for BBC Radio 2 Country.
