On November 24, 2010, Biraghi made his first team appearance in a competitive match against Twente in the 2010–11 UEFA Champions League, downing Goran Pandev. Inter had lost a number of first team players due to injury long before the match.

Biraghi will start in Inter's next game in the Champions League, with Davide Santon as the German team's right wing forward and Cristian Chivu unable to participate due to injury.

He was loaned to Juve Stabia, a Serie B club. In the club's first league game of the season, he made his debut against Empoli 2–1.

Biraghi was signed by Serie B club A.S. Cittadella on loan in summer 2012, with the ability to buy half of the registration rights. Cittadella used to pay €150,000 in June 2013. Inter also paid a €50,000 loan subsidy to Cittadella (Italian: premi di valorizzazione).

On September 2, 2013, Biraghi was loaned to Catania for €300,000.

Inter bought Biraghi from Cittadella for €610,000 (€600,000 more than Simone Pecorini's current registration rights). In 2014, he began a new four-year deal with Inter. However, he spent two seasons on loan with other clubs before he became a make-weight of another signing.

On July 6, 2014, he agreed to a two-year loan agreement with Chievo.

Biraghi and Granada CF, a La Liga team, signed to a one-year loan agreement on August 27, 2015.

Biraghi was sold to Pescara, a newcomer in a permanent contract on July 11, 2016. Pescara also signed Rey Manaj on loan, as well as Gianluca Caprari, who was sold to Inter but loaned back to Pescara for a season. At the end of the season, Pescara was relegated.

Biraghi joined Fiorentina on loan from August 15th, with the promise to buy the player.
