Grimes has appeared in Both the Boys Love Mandy Lane, War Eagle, Arkansas, and the assassination of a High School President, which received a limited release in March 2009. As Ryan Lafferty, the illegitimate son of William Walker, and patriarch of the show's family, he appeared in ABC drama Brothers & Sisters. He became a series regular starting with Season 4.

He appeared in the 2010 FX film pilot "Outlaw Country" with Haley Bennett and Mary Steenburgen. Eli Larkin, a cowboy, was a member of Grimes. The pilot was not picked up by FX, but it did air on August 23, 2012 as a television show. In 2012, he appeared in Liam Neeson's thriller Taken 2 as Neeson's daughter's boyfriend.

During the show's sixth season, Grimes also portrayed James in True Blood for six episodes, but later left the role. According to reports, he resigned because he refused to participate in same-sex kissing or sex scenes or to play a character that was attracted to males.
