Yamazaki made his acting debut in 2010 when he was cast as a mysterious high school student in the television series Atami no Sousakan. That was followed by a supporting role in the TV series Clone Baby, in which he played a hacker.

Control Tower, Yamazaki's first film, was released in April 2011. In the film, he sang and played the guitar, and his acting was well-received. He appeared in The Wings of the Kirin (2012) and as the lead in the horror thriller film Another, as the second time by Control Tower co-star Ai Hashimoto. He appeared in the following films: The Chasing World 3 and Live action of Love for Beginners. Sato Family Breakfast, a gay-themed film that aired on television (BS Japan), was a 2013 production. In 2015, he appeared at the 2nd Taiwan International Queer Film Festival (TIQFF). Besides that, he appeared in JINX as a supporting actor. and Koukousei's 35-sai no Koukousei. He starred in L DK, a film based on the highly acclaimed manga of the same name, alongside Ayame Goriki in 2014. The film was a success in his career, and it attracted more national attention. He appeared in three drama series, including Baseball Brainiacs, in which he was co-starring Sota Fukushi and Yuto Nakajima. In addition, he made his stage debut in Satomi Hakkenden as the main lead Inuzuka Shino. In 2015, he appeared in a film adaptation of a famous manga titled Heroine Shikkaku alongside Mirei Kiritani. He appeared in Orange alongside Tao Tsuchiya. The film was a huge success, grossing nearly US$28 million and becoming the ninth highest-grossing Japanese films of 2016. He received the 39th Japan Academy Award for Young People of the Year from those two films, as well as being nominated at the 2016 Hochi Film Awards. He appeared in the television adaptation of the legendary manga Death Note as L and the NHK Asadora Mare, which was led by the aforementioned Tsuchiya Tao earlier this year.

Yamazaki appeared in Your Lie in April 2016, as Kosei Arima, a gifted pianist, alongside Suzu Hirose, who played Kaori Miyazono. Yamazaki had to learn the piano and trained for six months before beginning to film. He appeared in Wolf Girl and Black Prince's live action film and the television series A Girl & Three Sweethearts. Yamazaki was also selected to appear in a special short film to commemorate Yasu Hara's historical manga Kingdom as the main protagonist, Shin. He reprised his role in the live action feature film, which was released on April 19, 2019. In 2017, he co-starred with Haruna Kawaguchi in One Week Friends. Critics largely praised Yamazaki for successfully portraying Yuki Hase, the cheerful and friendly Yuki Hase. He appeared in a few more live action films; JoJo's Bizarre Adventure: Diamond Is Unbreakable Chapter I, Psychic Kusuo's Deaffection Saiki K. and Hirono's Forbidden Secrets. He appeared in Rikuoh's TV series towards the end of 2017.

Yamazaki was the lead role for a top nightclub host in the NTV winter drama Kiss that Kills, his first lead role in a drama starring Mugi Kadowaki, Mackenyu, and Masaki Suda in January 2018. His film A Forest of Wool and Steel was also released, in which he portrayed a piano tuner. Yamazaki spent time in Hokkaido to learn and practice piano tuning. The Emperor and Empress of Japan attended a special screening of the film in which they interacted with the actor. Yamazaki appeared in the television series Good Doctor in the summer of 2018. It tells the tale of a young autistic man who becomes a pediatric surgeon based on the 2013 Korean drama of the same name. The drama received a warm response from viewers, and it was the first time he acted as a doctor and assumed a more unconventional role. It earned him the Television Drama Academy Award for Best Actor.

Yamazaki went on to star in Wotakoi's live action film Love is Strange for Otaku in 2020. In February 2020, the film was released. Hirotaka Nifuji, a aloof working adult who is a huge fan of video games and Narumi Momose, who was played by Mitsuki Takahata, was the principal role. In Netflix's interpretation of Alice in Borderland, he is best known for his leading role as Ryohei Arisu. The series is expected to air for two seasons.
