He spent his first professional work on stage, appearing in Good Rockin' Tonight, a musical based on TV producer Jack Good's life.

Four BBC period dramas include The Moonstone with Keeley Hawes, The Buccaneers with Carla Gugino, Madame Bovary with Frances O'Connor, And in the 2007 five-part series Cranford, Sir Charles Maulver. He appeared in ITV's seven-part drama Wonderful You in 1999 with his future mother-in-law Phyllida Law and future brother-in-law Richard Lumsden. For The Carte Noire Readers, Thomas Hardy's Tess of the d'Urbervilles to Kiran Desai's The Inheritance of Loss was shot in a series of readings of love scenes from a collection of classic and modern love tales. In 2011, he appeared in Honeymoon for One, starring Nicollette Sheridan.

He is also the producer of The Song of Lunch, a 2010 BBC/Masterpiece film starring Emma Thompson and Alan Rickman.

Greg Wise appeared in Brad Fraser's Kill Me Now at the Park Theatre in Finsbury Park, London, in February 2015. In the BBC's two-part television adaptation of Sadie Jones' debut novel The Outcast, Wise played emotionally distant father Gilbert Aldridge in July 2015. In series 1 and 2 of The Crown, Wise portrayed Lord Louis Mountbatten.

In 2019, Wise defeated the celebrity version of The Great British Bake Off to raise money for Stand Up to Cancer.

Wise was a contestant on the nineteenth season of Strictly Come Dancing in 2021. In the fourth week, he was partnered with Karen Hauer and the pair were forced to end.
