He earned the Pennsylvania state light middleweight championship after a 10th round unanimous decision over opponent Keenan Collins later this year. Rosado defeated Sechew Powell in the 9th round of the vacant WBO InterContinental light middleweight crown on June 1, 2012.
He defeated undefeated WBA and IBO middleweight champion Gennady Golovkin on January 19, 2013 at The Theater in Madison Square Garden, New York City, after amassing a record of 21-5 in 26 pro fights. In the second round, Golovkin dominated the fight, inflicting Rosado with a nasty bruise near his left eye. When Rosado was struck by right hands and strong jabs from his foe, he bled from the nose. Billy Briscoe, his brother, put an end to the contest in the seventh round.
On October 26, 2013, Rosado defeated undefeated Peter Quillin for his WBO middleweight title. In the tenth round, a nasty cut was made on Rosado's left eyelid, causing a doctor suspension and winning Quillin the technical knockout match. Despite numerous commentators' arguing that the contest was not fair, the judges had scored the match 87-83, 89-81, and 90-80 in favor of Quillin at the time of the suspension. Rosado yelled "bullshit" at the Showtime commentary team as soon as the combat was over, telling Jim Gray that he wanted a rematch; a rematch never materialized.
Rosado won the WBA Continental Americas and WBO International super middleweight titles in 2013, defeating former IBF welterweight champion Joshua Clottey in 2015 and knocking out highly rated undefeated opponent Bektemir Melikuziev in 2021.
