Club career
Rugani, a boy from Lucca, Italy, began his football career with Empoli's Tuscan side in 2000, at the age of 6. He stayed in the club's youth academy for 12 years before being transferred to Juventus, a Serie A club on loan in August 2012 for €150,000. Rugani joined Juventus' Primavera (under 19) youth squad, where he was a regular starter in the team from 2012 to 2013, winning the Coppa Italia Primavera.
Rugani was bought by the club on 31 July 2013 in a co-ownership contract for €500,000 and then sent back to Empoli on a season-long loan agreement ahead of their 2013-14 Serie B campaign.
Rugani, Empoli's star defender during their productive season, had made 40 appearances and scored two goals for the club as a result of his age. In a 4–0 home win over Reggina, he scored his first career goal off a corner on March 22. The club finished the season in second place, gaining automatic promotion to Serie A, alongside the champions Palermo.

Rugani has officially returned to Juventus in the summer of 2015. He made his club debut in September 2015 when he came as a replacement in a 2–0 home victory over Sevilla in the UEFA Champions League group stage, while also making his European debut in the process. In the round of 16 of the Coppa Italia, he made his debut with the club on December 16th in a 4–0 victory over Torino, a cross-city rival, in the Derby della Mole.
