After entering Johnny & Associates at the age of ten, Yamada began his career as an entertainer. His mother, a fan of KinKi Kids, applied for an audition that sought to select future male idols. The audition took place in the summer of 2004 and was broadcast in a segment of a Japanese television show Ya-ya-yah. In August, Yamada made his first television appearance.

He began performing as a backup dancer for Tackey & Tsubasa, NEWS, Kanjani8, and KAT-TUNE, and appeared on Shounen Club regularly since fall 2004.

He started acting after his success as a dancer. In 2006, he made his acting debut in Tantei Gakuen Q SP, as Ryu Amakusa.

He was chosen to be a member of a temporary group called Hey! in April 2007.

Shin Kibayashi, author of the Kindaichi series and Tantei Gakuen Q, and Yamada's Tantei Gakuen Q had envisioned a new Kindaichi series with Yamada for almost five years, but it was later revealed. When filming Tantei Gakuen Q in 2006 and 2007, they met on set. After filming the series, Kibayashi asked Yamada if he'd like to play Kindaichi in the future, because Yamada was only 14 years old at the time, too young to play the role of a high school student.

Ryosuke Yamada reprised his role as Kindaichi and appeared in another special titled Kindaichi Shonen no Jiken (The Prison School Murder Case).

After the specials, Ryosuke was invited to continue starring as Kindaichi for a new serial drama for the Kindaichi series. Kindaichi Shonen no Jikenbo N (neo) will be the new series, which will premiere in July 2014.

In the live-action movie version of the famous manga Assassination Classroom, Yamada made his movie debut as the hero, Shiota Nagisa. On its first week, the film was a big success, topping the box office of Japan.

JUMP was selected as the TV host of NTV's annual 24 Hour Television, which was a campaign to raise money for all the people going through difficult times, as well as V6 from Johnny's group. In the 24 Hour Television drama special called Okaasan, Ore Wa Daijoubu, Yamada and Yoshihiko Inoho were cast members. He portrayed a high school student who is passionate about soccer but who also suffers from a brain tumor. Yamada lost 6 kilograms in a week, just for the role. He was selected as host of the Jyanino channel in 2022, along with the other participants.

Yamada appeared in Grasshopper, another film that was released on November 9, with Toma Ikuta. On its first week, the movie debuted at number two. Yamada's role as a knife-wielding assassin in this film received raves, including from his veteran co-stars, Toma Ikuta, Tadanobu Asano, and Jun Murakami.

In the 39th Japan Academy Award's Newcomer of the Year category for his role as Nagisa Shiota in Assassination Classroom and Rookie actor of the Year in the Rookie category for his role in Semi in Grasshopper in early 2016.

It was announced on May 24, 2016, that Yamada would play Edward Elric in the Fullmetal Alchemist live-action film that was set to be released in 2017.

Yamada was leading off Fuji TV's gettingu 9 (prime-time slot)'s fall drama, including Cain and Abel. It first appeared in October 2016. It's Yamada's first drama with a romantic love theme, and it's also the first Heisei born from Johnny's to appear in a Getu 9. The tale is based on the Old Testament of the Bible, and it is also a remake of James Dean's classic film East of Eden. Yamada is a salaryman who falls in love with his brother's sister and is looking for his father's love. Yamada was runner-up for Best Leading Actor in the Autumn TV Drama Academy Award 2016 for his role in this drama. He was also the youngest person to be nominated in the lead actor category that season.

On December 2, 2016, Yamada was cast to lead a movie titled Miracles of the Namiya General Store, based on the best-selling book of the same name by an award-winning writer, Higashino Keigo. The novel was described as one of Higashino's finest works. It tells the story of two people from different eras (1980 to 2012), which will be linked by a letter, a tale depicting human bonds and miracles. Yamada will portray a delinquent who stumbles upon a cryptic store that leads to several events that change his life. The film will be directed by Ryuichi Hiroki, and is scheduled for release in Fall 2017.

Yamada's appearance in the Namiya General Store's "Miracles" received mainly favorable feedback. Nishida Toshiyuki, his co-star, praised Yamada's work, said he is the Japanese version of James Dean, and wrote a special letter to Yamada thanking his acting. At the 41st Japan Academy Awards, the film was nominated for six categories, including best picture and best director.

Both Namiya and Fullmetal Alchemist debuted in Japan's top 10 for their first weekend. Yamada's success as Atsuya and Edward Elric in Fullmetal Alchemist earns him the Newcomer Actor Award at the 91st Kinema Junpo Award, making him the second Johnny's to win the award after Ikuta Toma.

Yamada was the lead in the NTV 10p.m drama for the winter of 2018. The title Momikeshite Fuyu, a family entertainment drama that tells the tale of three influential siblings of a doctor, a lawyer, and a police officer who is struggling to conceal information relating to their relatives. Yamada is the youngest child, an elite Police Officer who graduated from the University of Tokyo, who has troubled to be accepted by his family amid the shadows of his older siblings. The series premiered on January 13, 2018, with a score of 13.3%.
