At Henley-on-Thames, Dexter was educated at Chiltern Edge School, King James' College (now Henley College), and the London Academy of Music and Dramatic Art. A Touch of Frost, Dalziel, Pascoe, and Families, as well as the long-running Night and Day, were among her many on-screen appearances. Dexter, a stage actor, has appeared in both straight plays and musicals in London's West End. She received the Olivier Award for Most Promising Newcomer in Theatre in 1987 for her role in Dalliance's 1986 revival. Nancy appeared in Sam Mendes' 1994 revival of Oliver! She appeared with the Royal Shakespeare Company and the Royal National Theatre in London for many years. She appeared in Bad Girls: The Musical at the Garrick Theatre, London in 2007, and can be seen on its original cast recording, as well as the London Palladium cast recording for Oliver! In the London Palladium's production of Sister Act from September to October 2010, she took over as Mother Superior.

In 2011, Tom Ellis, Sue Johnston, and Miranda Raison co-wrote and produced the three-part BBC comedy drama series Sugartown, starring Tom Ellis, Sue Johnston, and Miranda Raison. In Ian Rickson's Young Vic production of Hamlet, she was cast as Gertrude opposite Michael Sheen, previewing in October 2011. In The Lion the Witch and the Wardrobe at the Threesixty Theatre in Kensington Gardens until September 9, 2012, Dexter appeared as the White Witch in The Lion the Witch and the Wardrobe. Dexter appeared in Viva Forever, a teenage version of the Spice Girls in 2012. Selina Crow appeared in Father Brown episode 5.3 "The Eve of St John" on BBC TV. Faith Dingle appeared in Emmerdale as Faith Dingle in February 2017. She also appeared in The Attack of the Adult Babies, a comedy horror film, and in BBC TV series Poldark. Dexter will leave Emmerdale on October 7th, 2019. In the episode broadcast on October 25, 2019, Dexter's final scenes were reenacted; in 2021, it was revealed that Dexter would reprise her role as Faith.
