Radhika was sent by the Welham Girls' boarding school in Dehradun, Uttar Pradesh, in the 1960s. Prannoy Roy was a friend of Radhika during her teenage years. Prannoy was born in Calcutta and was sent to The Doon School, a boys'boarding school in Dehradun. Radhika and Prannoy migrated to London, United Kingdom for higher education, where they were married and then returned to India, settling in Delhi. She studied at the Oldfield Fleming School in London and became a qualified speech pathologist. Miranda House, University of Delhi, has also graduated with a degree in English literature.

Radhika Roy began her career as a journalist at The Indian Express, where she worked for the editing desk. She worked as a news coordinator for the India Today newspaper. Roy left her journalism to pursue a post-graduate degree in broadcast journalism at The New School For Social Research in New York, United States. At the New York University Tisch School of the Arts, she also applied for and completed a course in television production. Radhika Roy and her economist husband Prannoy Roy formed New Delhi Television Ltd in 1984 (NDTV). Both Roys are thought to be the company's founders, but Prannoy claims Radhika was the company's original founder and that he joined later. Doordarshan's public radio broadcaster, and the company became India's first independent news broadcaster. NDTV is considered a legacy brand in India, and it served as the foundation of broadcast journalism.

Radhika Roy, the managing director of NDTV from 1998 to 2011, before which she was chairman. She has also served as the CEO of a corporation. While Radhika Roy handled the editorial and backend operations, Prannoy Roy became the network's public face, while Radhika Roy handled the backend and production processes. She grew a reputation for demanding high standards of editorial integrity and impartiality. At a time when other broadcasters had none, Radhika had developed a legally binding code of conduct for journalistic ethics in the company. Roy has also been praised for her social responsibility and leadership, as she introduced sanitary napkins in NDTV offices at a time when the discussion about destigmatization of periods in workplaces had yet to enter public discourse. "I]f Prannoy Roy, the company's frontman, is its image, Radhika Roy is the organization's heart and soul," a senior executive said.

In a 5-year alliance with Star India, NDTV launched India's first 24x7 independent news channel. NDTV handled the editorial and production aspects of the deal in exchange for a fee with an escalation clause, while Star managed the infrastructure and retained the funds. The relationship was terminated in 2003 due to a lack of complete editorial control over NDTV. Following the split, NDTV became an independent news broadcaster after launching its own news channels NDTV 24x7 and NDTV India. In May 2004, the corporation went public and became the country's top media company in terms of market capitalization by the end of the year.

Since 2011, Radhika Roy and her partner Prannoy Roy were named as the executive co-chairpersons of NDTV. Radhika continued to serve as a steward for the company's editorial branch. According to reports, she was highly respected by her employees and remained heavily involved in the company's day-to-day activities to the point that she was recognized as the de facto CEO, although one former employee described her work style as often controlling because there was no effective decentralization in the organization. After Narendra Modi became Prime Minister, the government came under increasing government pressure through lawsuits and coercion of advertisers on the network. Following widespread demonstrations, the government attempted to block the Hindi news channel NDTV India in 2016 but it was forced to retract. After a NDTV news presenter challenged statements made by a ruling party spokesperson, the Central Bureau of Investigation raided the Roys' headquarters and the Roys' residence in 2017.

Radhika and Prannoy Roy had been barred from serving as board or board positions in the company for a period of two years, starting in June 2019. The order was appealed and postponed by the Securities Appellate Tribunal (SAT). SEBI issued a fine on the Roys worth 270 crore (US$3.4 million) in December 2020. The Roys were ordered by SAT to deposit half of the money as conditional to a second hearing. The company also applied to the Supreme Court of India (SCI), which exempted them from the deposits. Justice Dhananjaya remarked that it had been "brash" for the tribunal to order it.

Doubts had also arisen by 2015 after the Roys had a controlling stake in a debt pact with billionaire Mukesh Ambani's Conglomerate Reliance Industries following a string of loan restructurings necessitated by the NDTV's downturn due to the global financial crisis. According to a survey published in late 2019, Radhika Roy directly owned 16.32% of the company, while her husband held 15.9 percent of the shares. Through a 50:50 holding company called RRPR Holding Pvt Ltd, the two companies acquired an additional 29.18% interest.
