Lemon began acting with The Rocks Players in inner city Sydney, Leichhardt, before graduating from the University of New South Wales in 1982. After working with Rodney Fisher's musical Steaming, her professional career began in 1982 with a small appearance in Sons and Daughters and then, in the eighth season of The Young Doctors, Naomi Zelda Baker appeared as Nurse Zelda Baker. Marlene Warren appeared in the television series Prisoner from 1984 to 1985, receiving critical acclaim and a gold medal for her performance.

Lemon appeared in numerous Australian musicals between 1985 and 1988, performing for the Sydney Theatre Company, Hunter Valley Theatre Company, and Northside Theatre Company, under the artistic direction of Noel Ferrier, Jon Ewing, Jean-Pierre Mignon, and Rodney Fisher. Lemon appeared in Judy Morris' film Luigi's Ladies in 1989, and was also cast as the lead actress in Jane Campion's film Sweetie. The latter role earned her a nomination in the AACTA Awards for Best Actress in a Leading Role, and she also received the AFCA Awards in the same category.

Lemon appeared in the television series The Flying Doctors and G.P. between 1991 and 1992 as Brenda Riley. She appeared in numerous theatre productions, including Summer Rain by Rodney Fisher, critically acclaimed The Venetian Twins by John Bell, and Robyn Nevin's The Girl Who Saves Everything.

In 2021, Lemon appeared in Fiona Palmer's recurring role in Eden, Australia's television series Eden, and as a tourist in The Tourist, a critical acclaim series. The film Here Out West, which was starring Nancy, was released at the Sydney Film Festival in November 2021. Lemon appeared as Mrs Thornhill in the Sydney Lyric Theatre's theatrical version of North by Northwest from March to April 2022.
