In an editorial shot by Bruce Weber for Vogue Italia alongside model Lady Eloise Anson, Delevingne had her first modelling job at age ten. In 2009, she joined Storm Management. She worked in the industry for a year before deciding on a salary and going through two seasons of casting before presenting her first runway show.

When working part-time in the London office of a fashion website, Burberry's Christopher Bailey scouted Delevingne in 2012. Bailey's spring/summer 2011 campaign starred her.

Delevingne's first catwalk appearance at London Fashion Week in February 2011 was in February at Burberry Prorsum A/W collection. Burberry Prorsum S/S 2012 collection was launched and sold later that year.

She began the 2012 season by walking in the Grand Palais' Chanel Haute-Couture spring show.

Delevingne appeared in nine shows for fashion companies, including Jason Wu, Rag & Bone, Thakoon, Donna Karan, Tory Burch, Oscar de la Renta, and Carolina Herrera. She walked exclusively for the Burberry Prorsum Womenswear A/W 2012 exhibition at London Fashion Week. During Milan Fashion Week, she walked for Fendi, Trussardi, Moschino, Blumarine, DSquared2 and Dolce & Gabbana.

During Nina Ricci's last week of Fashion Week, she appeared in seven shows for Nina Ricci, Sonia Rykiel, Cacharel, Stella McCartney, Paul & Joe, Kenzo and Chanel. This was the start of what would be a long-term professional relationship with Chanel's lead designer and creative director, Karl Lagerfeld. "Cara is different," Lagerfeld said in an interview with Elle after the show. She's full of life and bursting with pep. I like girls to be wild, but at the same time, she is very beautiful and very funny."

In September 2012, the international Fashion Month debuted spring/summer collections for 2013. Marc Jacobs, Oscar de la Renta, and Marchesa were among the designers on show during New York Fashion Week. She walked exclusively for Burberry in London. She followed this appearance by walking in shows for Sass & Bide, Issa, Preen, Topshop, Mary Katrantzou, Giles, and Matthew Williamson.

She walked for Moschino, Emilio Pucci, Ermanno Scervino, Ports 1961, Anteprima, Iceberg, Gabriele Colangelo, Anthony Vaccarello, and Just Cavalli at Milan Fashion Week. She walked for Cacharel, Louis Vuitton, and Costume National during Paris Fashion Week. She had appeared in over 50 shows this season. Delevingne then proceeded to appear in the Victoria's Secret Fashion Show on November 7, 2012. She appeared in the 'Pink is US' segment and walked around Justin Bieber's appearance on 'Beauty And A Beat.' On December 4, 2012, the program premiered on December 4, 2012.

She appeared in the Chanel pre-fall 2013 exhibition in Scotland with the runway around Linlithgow Palace's courtyard. The collection, which was created by Karl Lagerfeld, was named "Paris-Edimbourg" and was influenced by Scottish style by using tartan fabrics. The show reignited national interest in the prospect of rebuilding the palace's roof. She gives the film a sense of vitality that is desperately lacking from the rest of it.

She appeared in a sketch for The Feeling Nuts Comedy Night, an annual 90-minute comedy television show that was aimed at raising testicular cancer awareness on October 24, 2014. She appeared in Die Antwoord's music video for their song Ugly Boy a month later. In December 2014, Delevingne appeared in Reincarnation, a short film directed by Karl Lagerfeld for Chanel starring Pharrell Williams and Geraldine Chaplin.

Fendi's F/W 2014 collection for Karl Lagerfeld opened in Milan before modelling for Stella McCartney in Paris. Delevingne went from walking in 'just about every single show out there' to only walking in the Marc Jacobs exhibition in 2013. "Will Cara Delevingne Walk in New York Fashion Week," news outlets reported. In the run-up to the fashion week, the designer was a celebrity.

Delevingne appeared in a Burberry fragrance campaign alongside British supermodel Kate Moss.

She opened and closed Topshop's 'Unique S/S 15' collection in London before heading to Milan, where she also opened the Fendi S/S 15 exhibition. Stella McCartney attended four shows for Givenchy, Yves Saint Laurent, Chanel, and Stella McCartney in Paris. She appeared in the Metiers d'Art Pre-Fall 2015/2016 collection exhibit in 2014. In the last episode of Playhouse Presents, Delevingne made her television debut opposite veteran British actress Sylvia Syms in June 2014. This was the first in a series of self-contained TV plays developed by British broadcaster Sky Arts.

Delevingne opened and closed the Chanel Haute Couture Spring 2014 exhibition in January 2014. In the A/W 2014 series, she showcases how she went on to close Burberry's show during London Fashion Week and debut in fellow British designer Giles' collection.

Several people were surprised that Delevingne had gotten more selective with the shows she appeared on. Delevingne appeared in ASAP Ferg's music video for his song Dope Walk in February 2015. Later in May 2015, she appeared in Taylor Swift's hit "Bad Blood" music video, which went on to smash Vevo's 24-hour viewing record, with 20.1 million viewers in its day of release.

Margo Roth Spiegelman co-starred in John Green's adaptation of Paper Towns (2015). On the first weekend in the United States, the film debuted at $6,650,140, placing it 6th at the box office. "Paper Towns isn't as deep or moving as it wants to be, yet it's also earnest, well-acted, and thoughtful enough to guarantee a spot in the hearts of teenage filmgoers of all ages," Rotten Tomatoes' critical consensus states. Justin Chang of Variety called Delevingne "the true find of the film," meaning that "on the evidence of her presence here, this striking actress is here to stay."

In Pan, a 2015 fantasy film, she played a mermaid. In Suicide Squad, an action film based on the DC comic book series of the same name, Delevingne co-starred Enchantress, a villainess with magical abilities. Despite a box office admonition, the film was released to largely critical feedback, but it was a box office hit. Critics generally gave Delevingne's result a mixed score.

In 2016, the film Kids in Love, a British coming of age drama, was released. Many up-and-coming young actors appear in the film, with Will Poulter and Alma Jodorowsky playing the role of Viola.

In the Luc Besson film Valerian and the City of a Thousand Planets, an extension of the comic book series Valérian and Laureline, Delevingne appeared with Dane DeHaan and Rihanna. The film debuted in December 2015 and was released in July 2017.

In the period romance film Tulip Fever, Delevingne plays Annetje. Kath Talent is also portrayed in the film London Fields, which is based on Martin Amis' 1989 book of the same name.

Delevingne appears in the female lead role as Vignette Stonemoss, a faerie war refugee in Amazon's Prime Video fantasy steampunk series. In 2022, she appeared in Only Murders in the Building for the second time.
