College career
From 2011 to 2014, Amos played at Penn State. He appeared in all 13 games as a true freshman and on special teams as a cornerback and on special teams. He had seven tackles and an intercept, as well as an intercept. Amos started all 12 games and had 44 tackles and two interceptions as a sophomore in 2012. His 40-yard dash time was seventh among all safeties, with his 4.03s time in the three-cone drill ranked second in second place among the best in the world. In the 20-yard short shuttle and the 60-yard shuttle, he also finished in the top tennies at the combine. Amos, together with Mike Hull, Donovan Smith, Jesse James, Sam Ficken, Miles Dieffenbach, Deion Barnes, and three others, decided to attend Penn State's pro day on March 19, 2015. He wanted to do the bench press, 40-yard dash, and positional drills for the team's representatives and scouts from 19 NFL teams in attendance. He had an excellent show overall and cut his time in the 40-yard dash from 4.56 seconds to 4.39 seconds, a new record. In the fifth round (142nd overall) of the 2015 NFL Draft, the Chicago Bears selected Amos. Brandon Marshall, a wide receiver for the New York Jets, had previously traded pick No. 2 by Brandon Marshall to the New York Giants. He was the eighth safety pick during the draft in 2016 and was only one of three Penn State players to be drafted in 2016, alongside Donovan Smith (second round, 34th overall) and Jesse James (fifth round, 160th overall).

During training camp, he competed with Brock Vereen and Sherrod Martin for the unfilled free safety position left after Chris Conte's departure from the free agency. In the season finale against the Minnesota Vikings on January 6, 2018, Amos made a season-high eleven tackles.
Pro Football Focus named their 2017 All-Pro squad on January 4, 2018, with Amos and Smith joining Amos at safety. He made ten starts in thirteen games, three passes tackled, one intercepted, two forced fumbles (including a return), and a touchdown.
