Early life
Notaro was born in Jackson, Mississippi, the niece of Mathilde "Susie" O'Callaghan and Pat Notaro. Her mother was born in New Orleans. Notaro was not born in Pass Christian, Mississippi, until kindergarten. Her family later moved to Spring, Texas, a suburb of Houston. Renaud Notaro, a year older brother, appears on a radio talk show host, and she also works as a year-old host. She was referred to as "tig" by her brother when she was two years old.
John Fitzpatrick, the mayor of New Orleans from 1892 to 1896, was Notaro's maternal great-grandfather. Notaro discovered that she is also a distant cousin of Gloria Steinem when attending season 5 of Finding Your Roots with Henry Louis Gates, Jr.
In a Mother Jones magazine interview, she said she looed school. Notaro dropped out of high school three years after failing three grades. Notaro obtained her general equivalency certificate in 1990 while living in Texas.

Personal life
On the set of the film In a World... Notaro met her wife, Stephanie Allynne. They got engaged on January 1, 2015 and were married on October 24, 2015. In 2016, they welcomed twin sons who were born via a surrogate using Allyne's eggs.
On July 30, 2012, Notaro was diagnosed with breast cancer in both breasts. During her live stage show at Largo in Los Angeles on August 3, she spoke about her cancer diagnosis and other personal challenges. Many comedians have lauded her performance, with several comedians lauding her work.

Career
Notaro, a lifelong music fan, migrated to Denver, Colorado, where she became involved in the music industry. In the mid-1990s, she became a band manager, working under the name Tignation Promotions.
Notaro's work promoting bands led her to Los Angeles, where she attempted stand-up for the first time in the late 1990s. Notaro has since been featured on Comedy Central Presents and the Sarah Silverman Show as a lesbian police officer. She and writer Kyle Dunnigan often collaborate. She co-hosted Professor Blastoff from May 2011 to July 2015.
Good One, her debut stand-up album, was released in 2011. Live is her 2012 album, which is a recording of a stand-up set performed shortly after she was diagnosed with breast cancer.
Notaro appeared on Conan in 2012 and on May of that year, it was broadcast to theaters around the country and on radio in edited form. She was more distant and observational in the past, but now she focuses more on her childhood and her life.
