Professional career

Printezis was loaned by Olympiacos to Olympia Larissa in the Greek League in 2006, where he averaged 11.5 points and 3.9 rebounds per game. He won the slam dunk competition at the Greek All-Star Game Weekend of the season. He was also voted Greece's Best Young Player at the end of the season. Following that success, he returned to Olympiacos.

After the second year, he signed a three-year deal with an NBA escape clause, earning €3.6 million euros with Unicaja Málaga, Spain's national ACB League team Unicaja Málaga.

After being transferred from Málaga, Printezis returned to his old club, Olympiacos, on April 27, 2011. He made a game-winning shot in the EuroLeague's 2011–12 season's Final, defeating CSKA Moscow, who had a few seconds remaining on the clock, to win the Olympiacos the EuroLeague championship.

He signed a new three-year contract with Olympiacos on July 12, 2012. He had considered a three-year, 4.5 million US dollars net income contract with Fenerbahçe Ülker before, but he decided to stick with Olympiacos instead. He also won the 2012-13 season championship of the EuroLeague with Olympiacos. He signed with Olympiacos until 2017.

Printezis signed a three-year contract extension with Olympiacos on June 14, 2017. On June 16, 2020, he signed a one-year contract with the team. Olympiacos announced on July 15, 2021, that Printezis had signed a new one-year contract extension.

After winning the Greek Basket League championship with Olympiacos, Printezis officially announced his retirement from professional basketball on June 17, 2022. He was the team's captain during his last season with the Reds.

The San Antonio Spurs selected Printezis in the second round of the 2007 NBA draft, making him the 58th pick overall. The printezis were then sold by the Toronto Raptors in exchange for a second round draft pick in the 2008 NBA draft, but the Toronto Raptors immediately traded Dragiz to the Phoenix Suns to draft future NBA All-Rookie Second Team member DeJuan Blair. Alexis Ajinça, a conditional second-round draft pick and cash, traded by the Toronto Raptors on January 20, 2011.

In a three-way trade, the paper rights to Printezis, along with Tyson Chandler and Ahmad Nivins' draft rights were sold to the New York Knicks on December 10, 2011. Andy Rautins from the Knicks and a second round pick from the Washington Wizards were included on the Mavericks. In addition to a Dallas 2012 second round pick, a 2013 Knicks second round pick, and cash considerations, the Wizards received Ronny Turiaf from the Knicks.

Printezis turned down a one-year contract with the Knicks in the summer of 2012, which was apparently less than the minimum NBA salary. In a contract that took Raymond Felton to New York, he and Kostas Papanikolaou's draft rights were sold to the Portland Trail Blazers on July 16, 2012. Eric Maynor was sent to the Portland Trail Blazers on February 21, 2013. In exchange for the draft rights to Sofoklis Schortsanitis, cash considerations, and a trade exception, he and Thabo Sefolosha were traded to the Atlanta Hawks on July 15, 2014, according to a trade exception. His draft rights, as well as a 2017 second-round pick, were sold to the Spurs in exchange for Tiago Splitter on July 9, 2015.
