Early life and high school career

Gillespie is the son of a police officer in the United States. He attended Archbishop Wood Catholic High School, where he was taught by John Mosco. Gillespie had no Division I scholarship offers coming into his senior year, but Rider, Hofstra, and Villanova had all expressed interest in a tournament, so he received offers from Rider, Hofstra, and Villanova. In a regular-season victory over Neumann Goretti featuring Kentucky recruit Quade Green, he scored 42 points. As a senior, he led the team to the PIAA Class 5A state championship in 2017. As a senior, Gillespie averaged 24.1 points per game and was named Philadelphia Player of the Year by the Philadelphia Daily News as well as Catholic League Most Valuable Player of the Year. On April 14, 2017, he signed a letter of intent with Villanova, joining Dhamir Cosby-Roundtree and Jermaine Samuels in the class of 2021.

College career

Gillespie enrolled at Villanova in the fall of 2017 and wanted to redshirt his freshman year at the school, but coach Jay Wright discouraged doing it because Gillespie "was playing above his years." In his second collegiate game, Gillespie scored in double figures, but he also had to adjust to the college game's quickness, which was soon integrated into the rotation. Due to a fractured bone in his left hand that was strained in December and January, Gillespie had to miss more than a month in December and January. In a win over Georgetown on January 17, the wing of the university returned to action. In a February 1 victory over Creighton due to Phil Booth's absence due to a similar hand injury. He had quality minutes and eight points, including two three-pointers. In an 86-75 win over Butler, Gillespie got off to a good start on February 10. In a victory over Radford in the first round of the NCAA Tournament, Gillespie scored nine points and had a steal. In the 79-62 NCAA championship game victory over Michigan, he had four points and five rebounds. In 14.4 minutes per game, he averaged 4.3 points per game. Gillespie's turnover ratio improved from 2.6 to -1, which Wright attributed to his teammates' involvement in helping them make them better.

In a 77–65 victory over Georgetown on February 3, 2019, Gillespie scored a season-high 30 points. In a 79-75 loss to Seton Hall on March 9, he had 22 points in a 79–75 loss. As a sophomore, Gillespie averaged 10.9 points per game.

Gillespie broke his nose in practice, requiring him to wear a mask for several games as he went into his junior season. In an 87-78 loss to Baylor in the Myrtle Beach Classic's championship game, Gillespie had 27 points and six assists. On November 25, he was selected to the All-Tournament Team and Big East Player of the week. In a 79-59 victory over St. John's, Gillespie had his first double-double with 17 points and a career-high 13 rebounds. In a 76-56 victory over Temple, Gillespie scored a season-high 29 points on February 16, defeating them by 76–56. Gillespie was named to the Second Team All-Big East at the conclusion of the regular season. On a team that finished 24–7, he averaged 15.1 points, 4.5 assists, and 1.2 steals per game.

Gillespie was named to the Bob Cousy Award watchlist as well as Preseason First Team All-Big East in his senior season. During a 72–60 victory over Creighton on March 3, 2021, he tore his MCL, snapping his season. Gillespie was named Big East Co-Player of the Year alongside teammate Jeremiah Robinson-Earl and Seton Hall's Sandro Mamukelashvili at the end of the season. He had 14 points and 4.6 assists per game. Due to the COVID-19 pandemic, Gillespie decided to take advantage of the extra season of eligibility offered by the NCAA.

Despite playing on a swollen ankle in an 89-84 victory at Providence, Gillespie scored a career-high 33 points on February 15, 2022.
