Tyson Griffin began his career in 2004 as a member of the Gladiator Challenge. In his third match, he fell to featherweight for a TKO win over emerging World Extreme Cagefighting featherweight champion Urijah Faber, which was Faber's first defeat in over three years. Griffin won all of his first six fights before meeting Duane Ludwig at Strikeforce: Revenge. In the first round, he defeated TKO, attracting major television audiences. Griffin decided to fight stateside after considering Pride Fighting Championship and UFC offers, and he's also joined the United Nations Football Championship so he could continue fighting stateside. Griffin had the opportunity to visit members of the Xtreme Couture camp for the first time while cornering for David Terrel at UFC 59 in Las Vegas.

Griffin made his UFC debut against David Lee at UFC 63. In the first round, he defeated the competition by a back-naked choke submission in the first round and later used the prize money to go to Las Vegas and join the Xtreme Couture camp. Griffin was branded a "gym jumper" by former teammate Nick Diaz, but Griffin denied the charge. At UFC 67, he started 2007 with a fight against Frank Edgar. Edgar was trapped in a tight kneebar, but the judges ultimately decided in his favor, awarding Griffin his first professional loss. At UFC 72 and UFC 76, respectively, he returned to action with two decision victories over Clay Guida and Thiago Tavares. All three of his bouts in 2007 were ranked in the top ten best of year.

Griffin defeated Gleison Tibau at UFC 81 and Marcus Aurélio at UFC 86 in 2008, winning unanimous decisions over him.

In a contest with title contention implications, he next met Sean Sherk at UFC 90. Griffin lost by unanimous vote in another three-round match, after a long battle.

Griffin beat Rafael dos Anjos at UFC Fight Night: Condit vs. Kampmann by unanimous decision.

In the second round of UFC 103, he defeated Hermes Franca via KO, his first victory in three years.

Griffin had intended to face Jim Miller at UFC 108 on January 2, 2010 but was forced to cancel due to a traumatic injury.

On June 12, 2010 at UFC 115, Griffin defeated Xtreme Couture training partner Evan Dunham. He was disqualified in the match due to a split decision.

On August 1, 2010, Griffin defeated former PRIDE Lightweight Champion Takanori Gomi, who was injured, in the match between Jones and Matyushenko. At 1:04 of the first round, Griffin lost via KO (punch). This was the first time Griffin had lost back to back fights, as well as the first time he had been denied entry in a fight after all of his previous losses had come to a conclusion. Griffin was clearly distraught with the death; he was pacing around the octagon and refusing to shake Gomi's hand.

Griffin was defeated by Nik Lentz by a split decision on November 20, 2010. Griffin had failed a post-fight drug screening, finding positive for marijuana following the match, according to the report.

In his featherweight debut on Versus 4, Griffin will meet Manvel Gamburyan in his featherweight debut on June 26, 2011. He won the fight by a majority decision.

Griffin lost via first-round KO on October 29, 2011, defeating Bart Palaszewski in the first round. Griffin had failed to meet the featherweight limit when weighing in, weighing in at 3 pounds over. Griffin was suspended 25% of his earnings, but the match took place at a catchweight of 148 lb. He wasn't announced until June 2012 that he had been suspended from the service right after the war.

Griffin also agreed to a 3-fight deal with the Resurrection Fighting Alliance (RFA).

In a lightweight match at Resurrection Fighting Alliance 4 on November 2, 2012, Tyson defeated TUF 8 champion Efrain Escudero. He gained the fight by a unanimous decision.

Griffin had signed a multi-fight contract with World Series of Combat in June 2013. Gesias Cavalcante, a Strikeforce veteran, met him on August 10, 2013. In the third round, he lost by TKO in the third round.

On June 21, 2014, Griffin defeated Luigiz Firmino at WSOF 10. He was disqualified by a majority vote in the fight.

Tyson Griffin returned to face former Bellator Jayne Bailey at URCC 34 on August 24, 2018, after a four-year absence/independence. He won an emotional victory by unanimous decision after his personal struggles during his time away from the sport.
