Early life
Glaser was born in Cincinnati, Ohio, on June 1, 1984, the niece of Julie E. Burke (née Burke) and Edward J. Glaser. Lauren is Lauren's older sister. She has ancestry from Germany and Ireland. Glaser spent the majority of her childhood in St. Louis, Missouri. She graduated from Kirkwood High School and briefly attended the University of Colorado Boulder before transferring to, and graduating from, the University of Kansas with a degree in English Literature.

Glaser is involved in a partnership with Not Safe, with Nikki Glaser co-creator Chris Convy. Both Convy and Glaser are residents of Greater St. Louis.

Career
At the age of 18, Glaser began doing stand-up. In college, she first jokes were written. In an interview with Rich Tupica in Revue magazine, she recalled it: "I remember it." I was a freshman in college, and everyone in my dorm ruled the cafeteria as like a study hall. I went to college and instead of studying, I just looked at them, judged them, and wondered, 'What will Sarah Silverman say about these people?' I didn't know how to write jokes; I just knew the stand-ups I knew. Since I didn't know what my perspective was yet, I wrote from the perspective of one of my favorite stand-ups.
Glaser appeared on The Tonight Show with Jay Leno, Conan, and two seasons of the reality show Last Comic Standing since then.
Perfect, her first stand-up special, was on Comedy Central on April 9, 2016. Bangin's second special, which debuted on Netflix on October 1, 2019.
