Duda made his major league debut against the Atlanta Braves on September 1, when he was facing starting pitcher Tommy Hanson, whom he met in high school. In three at-bats, he was hitless in three at-bats, but in the outfield, he made a "stellar sliding catch." Due to hamstring cramps, Duda had to leave the game in the eighth inning, which he explained after dehydrating on the plane from Buffalo to Atlanta. Duda batting against Hanson on September 17, 2010, batting for the first time in his career in major-leagues. Jerry Manuel, the former Mets manager, watched Duda during batting practice when he first was called up to the Majors, noting that Duda reminded him of Magglio Ordórz or Moisés Alou.

In Game 5 of the World Series, Duda's errant throw to home with 2 outs and the Mets leading 2–1 allowed Eric Hosmer to score the tying run, but the Mets lost the game when the Kansas City Royals defeated in 12 innings, completing the World Series championship.
