High school career

Marlowe, a senior, competed quarterback and safety on the football team at Holy Cross High School. He and Stanford wide receiver Devon Cajuste helped lead the Knights to the CHSAA Semi-final game in the state St. Anthony's by a score of 27–7. Holy Cross finished the season ranked 18th in the state. When he started with James Madison University, rivals gave him a two-star rating.

College career

Marlowe competed for the James Madison Dukes from 2011 to 2014. In 2010, he was redshirted. In 48 games played, he had 326 tackles, 12 interceptions, and 30 passes defensed. In 2012 and 2014, he was named First-Team All-CAA honors. In 2013, Marlowe received second-team All-CAA recognition, as well as third-team accolades in 2011.

Professional career

Following the 2015 NFL Draft, Marlowe was signed by the Carolina Panthers as an undrafted free agent. On September 5, 2015, he made the team's 53-man roster.

The Panthers of Marlowe played in Super Bowl 50 on February 7, 2016. He was out for the game as the Panthers lost by a score of 24–10.

Marlowe was put on the injured reserve on September 20, 2016, after suffering a hamstring injury.

The Panthers waived/injured Marlowe on August 8, 2017 and placed him on injured reserve. He was released on August 15, 2017.

Marlowe was signed to the Buffalo Bills' practice squad on December 5, 2017. On January 8, 2018, he signed a Bills reserve/future deal.

On September 1, 2018, Marlowe was waived by the Bills and was assigned to the practice squad the next day. On October 6, 2018, he was promoted to the active roster. On October 9, 2018, he was suspended and was re-signed to the practice squad. On December 12, 2018, he was welcomed back to the active roster.

Marlowe re-signed with the Bills on March 26, 2020. Following some roster moves, he was released as a part of the final roster cuts on September 5, 2020, but he re-signed the next day. Marlowe recovered a fumble against Cam Newton late in the fourth quarter to win by the Bills by 24-21. Marlowe was listed on the reserve/COVID-19 list by the team on November 14, 2020, and it was activated on November 19. During the 38-9 victory over Cam Newton against the New England Patriots on Monday Night Football, Marlowe got his first callout on the game in Week 16. Marlowe intercepted two passes kicked by Tua Tagovailoa during a 56–26 victory over the Miami Dolphins in Week 17.

On April 8, 2021, Marlowe signed with the Detroit Lions. He appeared in 16 games with nine starts, totaling 67 tackles and two passes defensed.

Marlowe signed a one-year deal with the Atlanta Falcons on March 31, 2022.

Following the Cowboys' 20-17 victory at SoFi Stadium in Inglewood, California, Chargers fans and a select few Cowboys supporters pushed, shoved, and grappled against each other while security battled to contain the increasing brutality. Judging by the replica jerseys worn by the veterans, the two teams in the skirmish were divided by their respective team loyalties. One Cowboys fan was seen throwing a right hook at a Chargers fan, only to have another Los Angeles fan drag him down and start yelling him. In addition, one Chargers fan had to be yanked by his belt from a Dallas supporter.
