Wojcicki joined Passport Capital, a San Francisco-based investment fund, and Investor AB after graduating. She was a health-care analyst for four years, focusing on biotechnology firms. Disillusioned by Wall Street's culture and its attitude towards health care, she decided not to take the Medical College Admission Exam to enroll in medical school and instead rely on biological research.

Wojcicki is best known as the co-founder and CEO of 23andMe, a direct-to-consumer DNA testing firm that allows consumers to test for ancestry and health risks. Linda Avey and Paul Cusenza founded the firm in 2006 with the intention of obtaining genomic information, which could include more details on diseases or treatments, particularly with the help of Glaxo and their $300 million investment. Wojcicki has expressed an interest in DNA testing, as it could provide consumers with enough information to identify potential genetic disorders.

Consumers can buy testing kits that contain ancestry, disease, and genetic variations. The company processes the genetic information in order to display the purchaser's online results.

The company is known for the discovery of 23 pairs of chromosomes in a normal human cell. In 2008, Time magazine named the company's personal genome testing kit "Invention of the Year." The Food and Drug Administration began authorizing 23andMe's health-related tests, including risks from cystic fibrosis, sickle cell anemia, some cancers, Alzheimer's, Parkinson's, and coeliac disease. 23andMe entered into a four-year partnership with GlaxoSmithKline in 2018 to produce new medications.

Wojcicki is a founder of the Xconomists, an ad hoc team of editorial advisors for the tech news and media company Xconomy. Wojcicki, the Most Daring CEO, was named by Fast Company in October 2013. She is a co-founder and board member of the Breakthrough Prize. She is ranked 93 on the Forbes list of the World's Most Influential Women as of 2020. Wojcicki joined the board of Cazoo in August 2021.

Wojcicki has been instrumental in Los Altos, California,'s downtown business district. Wojcicki and her then-husband Sergey Brin purchased at least half a dozen commercial buildings in downtown Los Altos in 2005. They sponsored activities and urban planning efforts throughout the downtown business district under the name of Passerelle Investment Company. In 2016, the company was renamed to Los Altos Community Investments and put a sharper focus on commercial real estate development. Los Altos Community Investments opened the State Street Market in downtown Los Altos in 2021.
