Playing career

After scoring 45 points in 46 games, Karlsson finished in 12th place in Allsvenskan scoring, the second-most on his team, in the 2011–12 season. With HV71, he made his senior Elitserien debut.

Karlsson signed a three-year, entry-level contract with the Anaheim Ducks, the team that drafted him in the second round, 53rd overall, in the 2011 NHL Entry Draft. For the 2013–14 season, he was on loan to HV71.

Karlsson made his NHL debut on October 11, 2014, scoring his first and second NHL goals against the Buffalo Sabres' Michal Neuvirth. On November 24, he was reassigned to the Norfolk Admirals, Anaheim's American Hockey League (AHL) affiliate. For the remainder of the season, Karlsson was reassigned between Norfolk and Anaheim but he ended the year in the AHL.

Karlsson was traded to the Columbus Blue Jackets in March 2015 (along with Rene Bourque and a second-round draft pick in 2015) in exchange for James Wisniewski and a third-round pick in 2015.

Karlsson was selected by the first-year expansion team, the Vegas Golden Knights, on June 21, 2017, after being "exposed" by Columbus for the 2017 NHL Expansion Draft. With the understanding that the Golden Knights would select Karlsson, the Blue Jackets traded a first-round pick, a second-round pick in 2019, and injured forward David Clarkson to Vegas.

Karlsson played for the 2017–18 season (Vegas' first in the NHL) centering a line with Reilly Smith and Jonathan Marchessault. All three players had a fruitful run, with all three putting up career-high offensive numbers. On December 31, 2017, Karlsson scored his first NHL hat-trick against the Toronto Maple Leafs, marking the first hat-trick in the Golden Knights history. On March 18, 2018, he scored his second straight NHL hat-trick against the Calgary Flames. Karlsson finished the regular season with 43 goals, a new NHL record for a player in an expansion team's inaugural season, in addition to providing 35 assists and a high plus-minus rating of +45. His 43 goals ranked third in NHL scoring, behind Patrik Laine (44) and Alexander Ovechkin (49). Karlsson was also named a finalist for the Lady Byng Memorial Trophy due to his high level of play and sportsmanship. He was crowned the winner of the trophy on June 20th, becoming the first player since Wayne Gretzky to win an end-of-season award in their team's inaugural season in the NHL. In the 2017-18 season, Karlsson was also named the best Swedish ice hockey player in the Guldpucken category.

Karlsson decided on a new one-year, $5.25 million deal with Vegas for the 2018–19 season following his career-high season. Karlsson said he was "betting on [him]self a little" and that he'd use the following season to show that his 2018-19 work "was no fluke."

Karlsson signed a new eight-year, $47.2 million deal with Vegas on June 24, 2019, having scored 24 goals and 32 assists (56 points) in the 2018-19-19 season. The deal goes into place from the 2019–2020 season to the end of the 2026–27 season.

During team captain Mark Stone's open air address, the center compelled supporters to yell, 'Go, Knights Go!' while taking off his shirt and throwing it into the crowd.
