Snipes was discovered by an agent when competing in a tournament at the age of 23. In the 1986 Goldie Hawn vehicle Wildcats, he made his film debut. Later this year, he appeared on the television show "Streetwise" as a drug-dealing pimp (first broadcasted December 5, 1986). In 1987, he appeared as Michael Jackson's nemesis in the Martin Scorsese-directed music video "Bad" and the Hollywood film Streets of Gold. Snipes was also considered for the role of Geordi La Forge in the television series Star Trek: The Next Generation in the same year, but LeVar Burton took the lead. Snipes auditioned and campaigned hard for Leroy Green's role in the 1985 cult classic film The Last Dragon, but the role was given to Taimak instead.
Spike Lee caught Snipes' appearance in the music film "Bad" on camera. Snipes scored a small part in Lee's Do the Right Thing for the bulk of Willie Mays Hayes' Major League debut, kicking off a string of box-office hits for Snipes. Snipes will appear in Mo' Better Blues and as the lead in Jungle Fever's interracial romance tale. Snipes was dubbed "the most celebrated new actor of the season" after Jungle Fever's success. Thomas Flanagan appeared in King of New York opposite Christopher Walken. In New Jack City, which was written specifically for him by Barry Michael Cooper, he played Nino Brown. In the 1994 film Sugar Hill, he also played a drug dealer.
Snipes has appeared in films including Passenger 57, Demolition Man (with Sylvester Stallone), Money Train, The Fan, and the United States. Marshals and Rising Sun, as well as comedies such as White Men Can't Jump, and To Wong Foo, Thank You for Everything! Julie Newmar, where he appeared as a drag queen. He received the Best Actor Volpi Cup at the 54th Venice Film Festival in 1997 for his appearance in New Line Cinema's One Night Stand. Snipes had his greatest commercial success with Blade, the title character, in 1998, grossing over $150 million worldwide. The film was the beginning of a franchise. He was also a member of the Hollywood Walk of Fame and an honorary doctorate in humanities and fine arts from his alma mater, SUNY/Purchase. Snipes reprised his role in Blade: Trinity, which he also produced, in 2004. In 2005, he sued New Line Cinema and David S. Goyer, the film's studio and director, respectively. He said that the studio did not pay him fully, that he was deliberately cut out of casting decisions, and that his character's screen time was reduced in favor of co-stars Ryan Reynolds and Jessica Biel. The lawsuit was later settled, but no details were released. Snipes later considered returning to Blade in the Marvel Cinematic Universe, but Mahershala Ali took over. Trinity was his last theatrical performance in the United States until 2010.
He appeared in The Contractor, filmed in Bulgaria and the United Kingdom, as well as Game of Death in 2012. Snipes had intended to appear in Spike Lee's 2008 war film Miracle at St. Anna, but he had to cancel due to tax issues; eventually, his role was replaced by Derek Luke.

Snipes and his brother formed the Royal Guard of Amen-Ra in the late 1990s, specialized in law enforcement and martial arts. Amen-Ra is also the name of his film company. Dr. John Henrik Clarke's first film, produced by Amen-Ra in 1996, was called A Great And Mighty Walk.
In 2000, the company was investigated for alleged links to the United Nuwaubian Nation of Moors. Snipes discovered 200 acres (0.81 km2) of land near their Tama-Re facility in Putnam County, Georgia, with the intention of purchasing and using it for his business academy. Both Snipes' company and the group used Egyptian motifs as their symbols. In the end, Snipes and his brother did not buy the land but instead established their company in Florida, Antigua, and Africa.
Snipes first started talks to defuse Fear Factor host Joe Rogan on Ultimate Fighting Match in 2005, but the deal fell through.
