Pascoe was a tour guide in London before she began her comedy career.

Pascoe has appeared in many television shows and panels, including Stand Up for The Week, The Thick of It, Mock the Week, Todd Margaret, The Increasingly Poor Decisions of Todd Margaret, Being Human, The Hundreds of Us, Can I Lie to You, Hypothetical, and W1A, as well as all-female sketch show Girl Friday (part of Channel 4's Comedy Showcase).

In 2007, Pascoe began doing stand-up comedy. Rachel Stubbings was runner-up in the Funny Women award in 2008 for the second year. Katherine Ryan received the award that year.

Sara Pascoe Vs. Her Ego was her first appearance at Edinburgh Festival Fringe in August 2010.

She appeared in episode 11 of the Comedian's Comedian Podcast hosted by Stuart Goldsmith and in Live at the Apollo in 2012. In 2013, she appeared on BBC TV's show QI.

Sex Power Money, her second book, was published in 2019, and it explores (mostly heterosexual) sexual relationships, with particular emphasis on male sexuality and sex work. Evolution biology and social science, as well as her own experiences and feelings, inform it. She also hosts a related podcast with the same name in which she interviews people who have had experience with sex work, stripping, and porn.

In 2016, she appeared on stage again with the show Animal. Taskmaster, a comedy challenge show, was broadcast on Dave in October and November 2016.

She has appeared in all three Frankie Boyle's Autopsy BBC shows (2014–2016), exploring social topics, and from 2017 to 2020, Frankie Boyle's New World Order has been a regular guest on his topical series Frankie Boyle's New World Order. In episode 1 of BBC series 2's episode 1, she also did a half-hour stand-up comedy.

On October 27, 2017, she appeared in an episode of the British travel documentary series Travel Man on Channel 4. Modern Monkey, a BBC Radio 4 series, began in February 2018.

She appeared as a panelist in two episodes of BBC Radio 4 panel show Just A Minute in April 2018. "Sara Pascoe vs Monogamy" was a BBC comedy short film released in May 2018.

In 2019, she was announced as the host of Dave's Giving Lectures, in which comedians gave a stand-up talk in the guise of a lecture. Dave revealed in February 2020 that they were commissioning two more movies, with series 2 being shot and broadcast in 2020 and series 3 in 2021. However, the Covid-19 pandemic caused the project to be postponed, and both series were shot at the National Gallery in London in summer 2021. In November 2021 and Series 3 in April 2022, there were two series 2 and three in April 2022.

On BBC2, she appeared in Travelling Blind in March 2019.

On BBC Two, a live recording of Pascoe's LadsLadsLads tour at the London Palladium was shown in April 2019.

Artificial intelligence found differences in brain activity that hadn't been previously seen by scientists, revealing 'hotspots' that can be used to distinguish the sexes. According to the results, there are key differences in the 'default mode network,' a brain device that aids us in processing the concept of'self,' and the striatum and limbic network, which are both important in learning and responding to rewards. The researchers, who came from Stanford University, said the report helps end a long-running debate over whether there are sex differences in the brain. However, previous studies had already pointed to differences in how men and women think, with women expected to have greater memory and writing skills, while men being more active and have better navigation skills (shown in diagram). In addition, it was already known that male brains were about ten percent larger than average, clocking in at 1.37 kilograms compared to women's 1.27 kilograms.
