Mancina has primarily worked as a composer for Hollywood soundtracks, such as his collaboration with Trevor Rabin on the Con Air soundtrack. He arranged several of the songs behind Disney's The Lion King (while Hans Zimmer wrote the orchestral score for the African chants with Nick Glennie-Smith and Lebo M.), as well as the Broadway musical. He produced the score for the thriller Twister (1996) and Bad Boys (1995). Mancina co-wrote several songs for Hanna-Barbera's 1990 animated film Jetsons: The Movie.

Mancina partnered with John Van Tongeren to write the theme for The Outer Limits' 1995 revival. Both of them appeared in ten episodes of the show's first season. Tarzan (for which he and Collins were recognized with a Grammy Award for Best Soundtrack Album) and Brother Bear collaborated with Phil Collins on two Disney animated feature films, as well as the one in which he and Collins received a Grammy Award for Best Soundtrack Album). Mancina created and arranged the sequence for "When You Wish Upon A Star" from 2006 to 2022 for the previous Walt Disney Pictures logo, which was co-arranged with and orchestrated by David Metzger. In addition, he created the music for the Disneynature logo.

Mancina made the music for the 2005–06 anime television series Blood+, which featured Hans Zimmer's music. He also wrote a score for Criminal Minds and Soldier Of Fortune, Inc., which was also on television.

Mancina has been involved in a variety of progressive rock projects. He toured with Rabin in favor of Can't Look Away and went on to produce tracks on the Yes album Union. He has also worked with Emerson, Lake & Palmer.

Mancina, a playwright, has written a musical based on the film August Rush, for which he had written the score. Mancina and Berger wrote the script for the musical, with Mancina composing the songs and Berger writing the book. In May 2019, the Paramount Theatre in Aurora, Illinois, under John Doyle's direction, made its world premiere.

Despite the fact that he did not appear in the 2020 film Bad Boys for Life, his original theme for Bad Boys is prominent throughout the film, as well as the end credits.
