Sharp's debut was in Alan Clarke's Rita, Sue and Bob Too (1986), starring Bob's wife, Michelle. Valerie in Jim Cartwright's acclaimed stageplay Road (1987) appeared in another Clarke-directed project. Clive Owen and Alan Rickman's supporting roles in The Rachel Papers (1989) and Stephen Poliakoff's Close My Eyes were among the actresses in The Rachel Papers (1989) and Stephen Poliakoff's Close My Eyes. Sharp was establishing herself as a talented actress and socially aware actress in Mike Leigh's Naked (1993) and the Jimmy McGovern-penned Priest (1994) who boosted her fame. She had become a well-known entertainer in the United Kingdom by the time she was in Prime Suspect 4: The Lost Child (1995) and The Full Monty (1997).

Although Sharp has appeared in a number of films over her career, she is certainly best known by television audiences. She was already playing lead roles in several well-written drama series by the late 1990s. Common As Muck (1997) was followed by Playing the Field (1998-2002), a story about a female football team with a five-series run. Sharp appeared in Great Expectations (1999), as Mrs Joe, and in Nature Boy (2000), as Martha Tyler, before deciding on Trudy Graham in Paul Abbott's BAFTA-awarded Clocking Off (2001–2003), which lasted four series. Russell T. Davies portrayed her opposite Alan Davies in Bob & Rose, resulting in a BAFTA nomination for Sharp. From Hell, starring Johnny Depp and Cheeky (1993), was preceded by another television drama written by Russell T. Davies in From Hell. "She killed God" in Stephen Baxter's role as played by Christopher Eccleston in The Second Coming (2003).

Sharp worked with Mike Leigh in Vera Drake (2004), which was followed by one-off television dramas including Planespotting, Born with Two Mothers, and Our Hidden Lives, which were all released in 2005. In ITV's supernatural drama series Afterlife, she appeared in the clairvoyant lead role of Alison Mundy opposite Andrew Lincoln's skeptical Robert Bridge. Despite the fact that the topic was seen as tense, critics and audiences alike generally greeted it positively. Sharp's work was highly praised, and she was nominated for a number of prestigious awards. In a This Morning television interview, she said that the guest stars – including Natalia Tena, David Threlfall, and Mark Benton – for the second series "were amazing."

In October 2005, after a ten-year absence from stagework, Emma in Sam Shepard's The God of Hell returned to the theater as Emma in Sam Shepard's The God of Hell at the Donmar Warehouse. Kathy Burke, a woman with whom she had previously competed for screen roles, directed her in what she described as "a black comedy about the poison at the heart of America." Lesley Sharp concentrated on theatre work for the next few years before re-appearing on television screens in 2008 in the three-part Lucy Gannon-penned drama The Children. She appeared with Russell T. Davies for the third time in 2008 as she appeared in the Doctor Who episode "Midnight." Sharp was later tipped to become the first woman to play the Doctor.

In early 2009, Sharp appeared in Petronella van Daan's latest adaptation of The Diary of Anne Frank on the BBC. In Channel 4's acclaimed drama series Red Riding, she played Paddy Considine's wife. Sharp appeared in The Rise and Fall of Little Voice at the Valiant Theatre in 2009, with Marc Warren and Diana Vickers, which ran from October to January. Sharp co-starred Janet Scott in ITV1's crime drama series Scott & Bailey from 2011 to 2016. Jan Starling appeared in the Sky1 comedy series Starlings in May 2012.

Sharp appeared in the three-part BBC series Capital based on John Lanchester's book of the same name in 2015.

Rosalind appeared in several episodes of Netflix's Fate: The Winx Saga as Rosalind, but she was dismissed from her role as Miranda Richardson after the first season.
