His first acting appearances came in Shakespeare's Many Ado About Nothing and Romeo and Juliet. When working odd jobs such as a book clerk and a video deliveryman, he began acting classes in Manhattan.

Patton's "big break" came in 2000 when he was cast to replace Steve Burns as Joe, the second host of the Nickelodeon children's television show Blue's Clues, after he was first introduced in the fall of 2000. Patton spent time in Williamsburg, Brooklyn, New York, as a waiter and a part-time actor. He was initially slated for a part of Blair Witch Project 2 at the time, but later on, he viewed the role as Joe, "a happy coincidence." Patton became a "household name" and has been described as "unqualified actor to the 5-and-under crowd and their families." Burns, who was involved in Patton's selection and helped prepare him, was subjected to "the same scrutiny" as he was. Out of 1,500 applicants, Patton was chosen. He had never seen the performance before his audition, but with preschool test audiences, Burns was the most popular. "I am positive that I got that part because of a Christopher Walken impression that I often do." There's a tape out there somewhere, with Steve Burns and myself doing a Blue's Clues scene as Christopher Walken, as we did at the screen test. I haven't seen the tape, but I know it exists somewhere. Burns trained Patton to take over the host position beginning in September 2000 and told him not to "think of [the show] as children's television" rather than "acting and telling the truth." Burns joined the show in January 2001, with the transition episodes airing in April 2002.

Patton's character was more like a preschooler and more innocent than Steve, according to Traci Paige Johnson, one of the designers and creators of Blue's Clues. Patton's portrayal as Joe brought him closer to the preschool audience, according to Angela Santomero, who created it. "Jump into a problem rather than figure it out first like Steve." Patton enjoyed being on Blue's Clues because, as he said, "I want to go to be a kid for twelve hours a day." By the time he joined the show, he called the show's crew "a real fun little family" and a "well-oiled machine." Patton said he had not fully grasped the immensity of replacing Burns, even after he became a parent and saw the show's effect on his own children. Patton, as well as Burns, appeared in the first episode of Blue's Clues & You!, where they played Joe and Steve to help their cousin Josh (which was played by Joshua Dela Cruz) learn how to care for Blue.

Patton appeared in short films Awake and Pharmboy in 2012, the latter of which was directed by his longtime buddy Lawrence Feeney. Mark Finkley, the director of Awake, was able to look beyond Patton's potential typecasting to a more believable role. In an audition for a new feature film, Patton impressed him. Patton was described as a "full [and] tremendous actor]" by Finton, who was both amusing on camera and exhilarating on stage. He appeared in the soap opera One Live to Live, in which he appeared in a departure from his previous roles as "a very unhappy man." He appeared in a small film shot mainly in New Jersey called Lies I Told My Little Sister, directed by William Stribling and starring Lucy Walters in 2014. In addition, he appears in Four for Fun, a Jason Cook film, alongside Jacob Young, Brytni Sarpi, and Annika Foster.
