In the West End's musical The Who's Tommy, Harmer made her stage debut at the age of six. She has appeared in a number of pantomimes, including Dolly Mixture (2004), the title role in Peter Pan, 2005, Alban Arena, 2006 in Worthing, and 2010 in High Wycombe), Dick Whittington (2007, Wolverhampton Grand Theatre), and the title role in Cinderella with Joe Tracini (2011, York Barbican). She appeared in Harry Potter and the Philosopher's Stone as an extra. Her big break came when she accepted the title role in CBBC's dramedy series The Story of Tracy Beaker, in which she appeared in five series, and in Tracy Beaker's sequel, Movie of Me.

My Family, Pie in the Sky, The 10th Kingdom, Beast, Touch Me, Karen Taylor, "Beep Beep" in the British television series The Beeps, as well as the pilot for Coming of Age. "Packers" appeared in the 2006 series of the Saturday morning show Mighty Truck of Stuff. She was one of the hostesses.

The Guardian announced in November 2008 that After You've Gone was to be cancelled by the BBC due to ITV's relocation of Coronation Street to the same slot on ITV1. In 2009, the BBC had announced a fourth series of the show in January of that year, but in 2008, the series came to a close.

Harmer signed a record-breaking deal with Universal/Decca worth £3 million under David Glatman's direction, some time before starting work on her debut album. In 2008, the album was recorded and written with the help of producer and songwriter Tom Nichols, and it was titled Superheroes. After being the lead singer of the TV show Dani's House, in which Harmer appears as the lead actor, the album's lead and only single "Free" was announced on May 25, 2009. According to the Official Chart, the song debuted at number 117. On the compilation album Pop Princesses 2009, "Free" was featured. The targeted release date of Superheroes was October 2009, which was later postponed to early 2010. The album had been cancelled by Harmer's record company due to poor sales of the lead single, according to her, "they are just waiting for the right time to release it." Harmer revealed on Twitter that she had left music because she wanted to concentrate on her acting career. The album never surfaced, but 30-second previews of each track and track list were included in early 2010.

In early 2010, Harmer returned to the role of Tracy Beaker in the 13-episode BBC series Tracy Beaker Returns, in which her character had developed into a care worker and had written an autobiography about what life is like for children in care. Tracy Beaker Returns was part of three series on television. It came to an end on March 23, 2012, with Harmer deciding to leave her job as Tracy. The Dumping Ground, a spin-off, was created, but she did not appear in two episodes at the end of season 6, but she did return for two episodes. Dani's Castle, a spin-off from Dani's House, followed her on television. This appeared on both 2013 and 2015 during the time of 2013 and 2015.

In fall 2012, Harmer was scheduled to appear in Disco Inferno, which was supposed to tour throughout the UK. However, it was reported on September 5, 2012, after an investor pulled out, that the musical had been cancelled. On the same day, Harmer revealed on Twitter that she will be a contestant in the forthcoming tenth season of Strictly Come Dancing.

In the 2013 UK tour of the Rocky Horror Show, Janet appeared.

Harmer appeared with 26 other celebrities in 2018 and performed the hit song Rock With Rudolph, written and produced by Grahame and Jack Corbyn. The song was released digitally on independent record label Saga Entertainment on October 30th, 2018 under the artist name The Celebs. The music video premiered exclusively with The Sun on November 29, 2018 and had its first television appearance on Good Morning Britain on November 30th. On the iTunes pop chart, the album debuted at number two.

Harmer would reprise Tracy Beaker's role in the 2021 CBBC film My Mum Tracy Beaker. Tracy is based on her friendship with her 12-year-old daughter Jess (Emma Davies), and it follows her as a single mother.

The actress, 34, is currently acting in Beauty and the Beast, but she was forced to stand in for her because she had been injured in a concussion. She took to her Story to tell her followers she was 'absolutely gutted' at the change of plans and promised to fans that she would be back as'soon as humanly possible.'
