The Chicago Sky, an expansion squad that year, selected Dupree 6th overall in the 2006 WNBA Draft. She was selected to the Eastern Conference All Star team as a rookie. Dawn Staley, her former college coach, was defeated in her first All-Star game. In addition to the Olympics, Dupree was also selected to the Eastern Conference All-Star Team in 2007 and 2009 (there was no WNBA All Star games in 2008). Dupree finished second in the WNBA with 268 total rebounds for the 2009 season.

Dupree was traded to the Phoenix Mercury in March 2010 as part of a multi-player, three-team contract. Dupree requested the trade.

In the last game of the season, Dupree had 24 points and six assists. Dupree was playing with Diana Taurasi and Brittney Griner during the 2014 season, when the Mercury had a record-breaking regular season record of 29–5 en route to the title.

Dupree was named a WNBA All-Star for the fifth time in her career in 2015, with an average of 14.1 ppg. The Mercury will return to the playoffs to defend their crown, but the Minnesota Lynx, who won the championship that year, swept the second round.

In 2017, Dupree was traded to the Indiana Fever as part of a three-team contract that earned Camille Little and Jillian Alleyne to the Mercury and the Connecticut Sun's 2017 first round pick, with Lynetta Kizer from the Fever as the eighth overall pick. During the 2017 season, Dupree will be voted into the 2017 WNBA All-Star Game for her sixth appearance in a row.
