Jensen studied Drama at Queen Margaret University in Edinburgh.

Clare Donnelly, the daughter of Glasgow criminal Jo-Jo Donnelly (played by Billy Connolly) in the 1993 BBC drama Down Among the Big Boys, her first television role was as Clare Donnelly, daughter of Glasgow criminal Jo-Jo Donnelly (played by Billy Connolly). In 1994, she appeared as Rosie McConnichy in the final series of BBC comedy shows for secretary Hilary (Rebecca Lacey), and Heather in Roughnecks, a BBC television series about workers on a North Sea oil platform. Jensen appeared on "Contact" in Dangerfield as the mother of a young girl with meningitis. Fiona Morris appeared in EastEnders as Fiona Morris. She appeared in the 2003 BBC drama Two Thousand Acres of Sky.

Jensen appeared in BBC Two/HBO's Extras as the socially inept Maggie Jacobs. At the 2005 British Comedy Awards, she received best television comedy actress and newcomer awards for her role on the first series. Jensen received two British Comedy Awards and a BAFTA nomination for her role in Extras in 2006. Emmy Award nomination for her role in the 2007 Christmas Special. Eleventh Hour, a short-lived drama series on ITV, starred her in January 2006. In A Cock and Bull Story later this year, she was Steve Coogan's agent. In 2007, she narrated the film Taking Liberties.

Jensen's first big role on American television came in September 2006, when she appeared as regular character Christina McKinney on ABC's Uncommon Heroes. Her character was born in the United States, but when she spoke to the show's creators, she loved Jensen's accent and changed the nationality of the character, keeping McKinney Scottish. Ugly Betty's production shifted to New York in mid-2008, causing a conflict for Jensen, who was unable to relocate from Los Angeles and dropped the show in 2009 near the end of the third season, but made a rare guest appearance in 2010 during the fourth season.

Jensen appeared in No Holds Bard, a BBC Scotland one-off special comedy on Burns Night in January 2009 as part of a line-up of special programs to celebrate Robert Burns' 250th birthday. She is the narrator of Channel 4's Embarrassing Illnesses and Embarrassing Bodies. She also narrated an advertisement campaign for Bank of Scotland and Persil detergent ads on television and radio. Olivia appeared on Purpose from September 2009 to May 2010. Jensen appeared in the 2009 BBC comedy Nativity! Martin Freeman played Jennifer Lore, a Hollywood producer's secretary and Paul Maddens' love interest.

She appeared on BBC comedy drama Accidental Farmer, playing a London executive who buys a derelict farm with her philandering boyfriend's credit card on December 21, 2010. Nanette the frog was the voice of Nanette the frog in the animated film Gnomeo & Juliet in 2011. In April of that year, she appeared in the ITV drama The Reckoning. She appeared on the BBC cookery programme Saturday Kitchen as a guest.

In the television film version of Agatha Raisin's M.C. Beaton novel Agatha Raisin and the Quiche of Death for Sky1, she played the eponymous lead character. She appeared in the eight-part series, which premiered on SkyOne on June 7, 2016.

Jensen appeared in Catastrophe, a Channel 4 comedy series, and in Susan Calman's BBC Radio 4 comedy series Sisters.

She appeared in a six-part TV series called Love, Lies & Records in 2017, depicting the gruesome life and entwined storylines in Leeds Registry Office. Jensen reunited with Gervais in the Netflix black comedy series After Life in March 2019, a role she retained for all three series. Debbie Dorell appeared in the second series of BBC TV drama Trust Me in April 2019. When an injured soldier discovers patients in his hospital ward's death, he investigates the enigmatic deaths. In addition, Jacqueline in Lady and the Tramp appeared in Jacqueline in 2019.
