Erin Boag, Du Beke's most reliable professional partner, is a member of the Society of Professionals. They met in 1997 and 1998, winning the 1999 and 1999 New Zealand Championships. They became professional in 2002 and became very popular in the United Kingdom. They won the IDTA Classic in Brighton in November 2003, the best on the circuit ever.

Since its inception, Du Beke has appeared in every series of BBC's Strict Come Dancing. He performed with singer Lesley Garrett in the first series and came in third place. He appeared on stage with actress Patsy Palmer for the third series and finished in fifth place. He performed with actor and impressionist Jan Ravens in the fourth series, but was disqualified in week 5 of the competition. Kate Garraway, the husband of the fifth series of the series, was voted out in week 7. Gillian Taylforth, his partner in the sixth series of the series, was voted out in week 2. In series seven, he was partnered with Laila Rouass. They made it to week 12 together.

He worked with Conservative politician Ann Widdecombe in series 8. Despite Widdecombe's poor dancing skills, the team made it to the quarter finals. The couple were canceled in week 5 for Anton's ninth series. Jerry Hall, a model and actor, appeared in the tenth series. In week 3, they were banned from returning to school. Fiona Fullerton, actor, was his companion for series 11. They were eliminated in week 8 in Blackpool. Judy Murray, tennis coach, appeared on the show's twelfth season. They were also banned in Blackpool on week 8. Du Beke and his co-worker Katie Derham were partnered on series 13 for Du Beke's first final in all 13 series and were disqualified in fourth place.

Du Beke was partnered with actress Lesley Joseph in a series 14. They were wiped out in week 5 of week 5. In series 15 in 2017, Du Beke was paired with television presenter Ruth Langsford. Anton was partnered with Fashion Journalist Susannah Constantine in 2018. They finished last (Du Beke's first time doing this) with an average of 12.00 (Du Beke's lowest). He was partnered with Emma Barton for the 17th series. Du Beke's first tens in all 17 series for their American Smooth were released in week nine; the pair's total number of fifteen tens was fifteen times during the series. They were joint runners-up with Karim Zeroual and Amy Dowden, but they were disqualified by Kelvin Fletcher and Oti Mabuse. In 2020, Du Beke was joined by former home secretary Jacqui Smith. They were disqualified in week 2 (first), Du Beke's second time finishing last.

Du Beke has also appeared on a few Children in Need and Christmas Specials of Strictly Come Dancing. He won the Children in Need 2008, a special dancing an American Smooth with Tess Daly in 2008. In the 2012 Children in Need special, he danced with Anne Widdecombe, but Russell Grant and Flavia Cacace took the lead. He did however win the Children in Need 2014 competition, where he and fellow dancer Natalie Lowe taught a group of children how to dance. For the 2004 and 2018 Christmas specials, Du Beke has also revived his collaborations with Lesley Garret and Ann Widdecombe. He has performed with Ronni Ancona, Su Pollard, Katy Brand, and Denise Lewis in the respective 2010 and 2016 Christmas specials. In 2015, Du Beke served as a judge on The People's Strictly, a one-off revival of the competition for the charity Comic Relief. In 2020, he reprised his role as a judge for weeks 4 to 5 to cover in for Motsi Mabuse, who needed to quarantine after travelling to Germany. Du Beke will be a permanent judge on Strictly Come Dancing from June 2021, replacing Bruno Tonioli, who appeared on June 24th, 2021.

After arriving at rehearsal with a spray tan, Du Beke said that his dance partner Laila Rouass "looked like a paki." Du Beke denied being racist, and said that the remark was made "in jest."

Du Beke was a contestant on the BBC Two show The Underdog Show in March 2007 and was paired with homeless dog Ginger. In week three of the season, they were disqualified.

Du Beke has hosted a cookery show Step Up to the Plate on BBC Radio. On the UK version of Hole in the Wall, which premiered on September 20, 2008 on BBC One, he served as both a captain and from series 2 as the host. He appeared in Victoria Wood's Midlife Christmas sketch "Beyond The Marigolds," where he spoofed his Strictly Come Dancing role by being paired with fictional soap actor "Bo Beaumont" (played by Julie Walters). When she was struggling to grasp Du Beke's warm up steps, Beaumont left the show.

Du Beke appeared on Channel 4's game show Draw It! during the week of 5 to 9 May 2014.

In Michael McIntyre's Big Christmas Show, Du Beke treated Craig Revel Horwood as a visitor on Christmas Day 2019.
