Wong tried stand-up comedy at the age of 23 after graduating from college. She hopped to New York City to pursue comedy and performed up to nine times a night.

Variety named her one of the "10 Comics to Watch" in 2011. She appeared on The Tonight Show, John Oliver's New York Stand Up Show, and Dave Attell's Comedian Comedy Underground Show right after. Are You There, Chelsea? was also cast as a series regular in the NBC comedy series Are You There, Chelsea? Chelsea has a late night show on Chelsea. In 2013, she appeared in VH1's Best Week Ever and MTV's Hey Girl. In addition, she appeared in Oliver Stone's Savages, opposite Benicio Del Toro and Salma Hayek, and as Kate in the film Dealin' with Idots.

Wong appeared in the ABC medical drama series Black Box in 2014, opposite Kelly Reilly and Vanessa Redgrave. Since then, she has appeared in many episodes of Inside Amy Schumer. Since 2014, Wong has been a writer on Fresh Off the Boat. Randall Park, the main cast member, had hoped for Wong for the writing role.

On Mother's Day 2016, Netflix released Baby Cobra, a stand-up special; the special was shot in September 2015, when Wong was seven months pregnant with her first child at the Neptune Theater in Seattle. "The special's appearance on Netflix unites the tastes of the uninhibited fans," New York Magazine says. During New York Fashion Week's Opening Ceremony on September 11, 2016, Wong spoke at, and walked the runway. Wong first appeared in the ABC sitcom American Housewife in October 2016. Hard Knock Wife, Wong's second Netflix special, was released on May 13, 2018. It was shot at the Winter Garden Theatre in Toronto in late September 2017, when she was seven months pregnant with her second child. In an episode of OK K.O., she portrayed Citrus Twisty, a soda genie. Let's Be Heroes.

In the 2019 Netflix film Always Be My Maybe, directed by Nahnatchka Khan and written by Wong, Park and Michael Golamco, Wong appeared with Randall Park. In the Netflix animated film Tuca & Bertie, Wong created Bertie, a fictional character.

Wong released a book on October 15, 2019, titled Dear Girls: Intimate Tales, Untold Secrets, and Tips for Living Your Best Life. She refers to it as a life guide for her two daughters to read when they're adults.

Don Wong, Wong's third Netflix stand-up special, debuted in February 2022. This is her first stand-up special in which she is not evidently pregnant.
